Laboratory-based project on the effect of MEK inhibition, through the use of Cobimetinib, on gene and protein expression in a cell line of Blast Phase Chronic Myeloid Leukemia. The K562 cells were treated for 48 hours with Imatinib IC50, Cobimetinib IC50 or a combination of the two before investigation changes in gene and protein expression in the MAPK pathway, other signalling cascades downstream of BCR-ABL1, apoptosis, and genes involved in proliferation. Overall, Imatinib demonstrated a predominant effect on post-translational processes, by affecting phosphorylation of molecules activated by the Phildelphia Chromosome. On the other hand, Cobimetinib influenced mostly transcriptional processes, and was shown to increase apoptosis and decrease proliferation in the K562 cell line. The combination of the two drugs does not seem to approvide additional benefit with respect to single arm treatments based on the results presented in this work.
